Engineering cell fate requires sustained expression of regulatory factors. But existing approaches often force a trade-off between transient RNA delivery and permanent genetic modification. Modified mRNA offers a powerful, integration-free way to express proteins. But expression is transient, often requiring repeated transfections to sustain cell-fate regulators or biosensors. Can we achieve sustained function without modifying the genome? 2/n
Enter self-amplifying RNA! saRNA contains machinery that amplifies RNA inside the cell, extending expression weeks beyond conventional mRNA delivery. By coupling saRNA with selection, we can establish cell populations that maintain useful transgene expression. 3/n
